Landry Jones attended the University of Oklahoma in 2008 and played for the Oklahoma Sooners football team from 2008 till 2012. In 2009, in the season opener match against Brigham Young, rookie Jones took over as the starting quarterback for the Oklahoma Sooners after Bradford suffered an injury to his shoulder, but unfortunately, Oklahoma Sooners went on to lose the game. 

However, in Jones next game which was his first as the starter on the team, Jones completed 18 of overall 32 passes for 286 yards along with three touchdowns as well as the interception in the Sooners 64–0 win over Idaho State.

In his second start of a college career, Jones threw a Sooner-record of six touchdown passes along with two interceptions in a 45–0 win against Tulsa. Moreover, in the same week, Jones was named AT&T All- America Player of the Week.

In 2010, Landry Jones continued his playing career as Oklahoma's quarterback and guided his team through its first four out-of-conference victories. 

In 2011, Landry Jones led the Sooners, credit to his dominant performances against opening foes such as Florida State and Tulsa. Moreover, he would outdo his 2010’s performance in the 2011 Red River Shootout by defeating Texas by a score of 55–17.

On January 5 of 2012, Landry announced that he was returning to his team Oklahoma Sooners for his final senior season. In 2012, Jones led the Oklahoma Sooners to a 10–2 record win with overall (8–1 in the Big 12), and Jones shared the Big 12 Conference Championship with Kansas State.

Landry Jones signed a four-year contract worth $2,599,220 with the Pittsburgh Steelers which included a $439,220 signing bonus and $439,220 guaranteed with an average annual salary of $649,805. In 2016, Jones earned a base salary of total $675,000.
